Compound Overview
ATIC-IN-1 inhibits dimerization of aminoimidazole carboxamide ribonucleotide transformylase/inosine monophosphate cyclohydrolase (ATIC), with a Ki of 685 nM. Because this dimerization is required for the enzyme's aminoimidazole carboxamide ribonucleotide (AICAR) transformylase activity, the compound reduces cell numbers and cell division rates, giving it anti-tumor activity[1]. It is supplied as a white to off-white solid (C21H33N7O5, MW 463.53).

For Research Use Only. Not for use in diagnostic or therapeutic procedures, and not for human or veterinary use. Handle in accordance with the Safety Data Sheet and your institution's chemical hygiene plan.
In Vitro
ATIC-IN-1 (10 μM, 50 μM) acts as a specific inhibitor and shows a nonspecific aggregation mechanism when 1 mg/mL and 10 mg/mL BSA are present[1]. At 100-500 μM for 48 h, it inhibits MCF-7 cell proliferation[1]. At 250 μM for 24-72 h, it reduces cell division rather than increasing cell death[1].
Cell Viability Assay[1]
| Cell Line | MCF-7 cells |
|---|---|
| Concentration | 100 μM, 250 μM, 500 μM |
| Incubation Time | 0 h, 24 h, 48 h, and 72 h |
| Result | Reduced the division rate to inhibit MCF-7 proliferation at 250 μM. |
