Compound Overview
ATR-IN-23 is a potent, selective ATR inhibitor, with an IC50 of 1.5 nM. It shows potent antiproliferative effects on LoVo cells and synthetic lethality on HT-29 cells, and can be used to study DNA damage response (DDR)-deficient cancers[1]. It has a molecular formula of C20H22N6O3S2 and a molecular weight of 458.56 g/mol.

Safety
For Research Use Only. Not for use in diagnostic or therapeutic procedures, and not for human or veterinary use. Handle in accordance with your institution's chemical hygiene plan.
In Vitro
ATR-IN-23 has significant inhibitory potency against ATR (IC50 = 1.5 nM) and strong antiproliferative activity against LoVo cells (IC50 = 0.073 μM)[1]. Against HT-29 cells, ATR-IN-23 shows moderate antiproliferative potency (IC50 = 0.161 μM)[1].
In Vivo
ATR-IN-23 causes acute toxicity at a maximum concentration of 2000 mg/kg and has moderate safety in ICR mice[1]. In BALB/c nude mice, ATR-IN-23 (50 mg/kg; once or twice daily; p.o.; 21 days) shows moderate antitumor efficacy[1].
| Animal Model | BALB/c nude mice[1] |
|---|---|
| Dosage | 50 mg/kg |
| Administration | p.o., once a day or twice a day for 21 consecutive days, dissolved in a solution of DMSO (10%), solutol (10%), and saline (80%) |
| Result | Exhibited moderate antitumor efficacy, with a tumor growth inhibition (TGI) value of 55% at dosages of 50 mg/kg twice a day. |
