Compound Overview
Azidocillin, a semi-synthetic Penicillin, is an orally active β-lactam antibiotic. It carries an azide functionality while retaining on-target activity within bacteria, and it has been studied for osteitis following dental surgery, otitis media, enterococcal septicemia, and other bacterial infectious diseases[1][2]. As a click chemistry reagent bearing an azide group, it can undergo copper-catalyzed azide-alkyne cycloaddition (CuAAc) with molecules containing alkyne groups, and it can also undergo strain-promoted alkyne-azide cycloaddition (SPAAC) with molecules containing DBCO or BCN groups. In Vitro
Azidocillin (0.004-16.0, >16.0 μg/mL; 24 h) suppresses growth of most bacteria isolated from human mandibular osteitis; mean MIC values run from 0.005 to 1.2 μg/mL depending on the genus, while just a few strains show MIC values above 2.0 μg/mL[1].
In Vivo
When Azidocillin (15 mg/kg; i.v.; single dose) is given intravenously to rats with bile fistulas and ligated renal pedicles, 12.9% of the dose is excreted in bile as biologically active drug over 1 hour; extensive biotransformation results in low levels of active drug relative to total excreted radioactivity[2].
