b-AP15 is a selective inhibitor of the deubiquitinating enzymes UCHL5 and USP14, which are associated with the 19S regulatory particle of the proteasome. By blocking these enzymes, b-AP15 disrupts the removal of ubiquitin chains from proteasome substrates, leading to accumulation of polyubiquitinated proteins and induction of apoptosis. This mechanism has been explored in cancer models, but also holds relevance in neurodegenerative disease research, where impaired proteasomal degradation and protein aggregation are hallmark features. b-AP15 provides a valuable tool for studying the role of deubiquitination in neuronal proteostasis and may inform therapeutic strategies targeting the ubiquitin-proteasome system in disorders such as ALS and Huntington’s disease.
