BIBW 2992 (Afatinib) is a potent and irreversible inhibitor of the ErbB family of receptors, including EGFR and HER2. Primarily developed for oncology applications, particularly in non-small cell lung cancer, its inhibition of EGFR signaling has broader implications. In the nervous system, EGFR signaling is involved in neurodevelopment, synaptic plasticity, and response to injury. Although BIBW 2992 is not widely used in neuroscience, its mechanism of action suggests potential utility in modulating neuroinflammatory responses and glial activation. Further research may uncover its relevance in neurodegenerative disease models where aberrant EGFR signaling contributes to pathology.
