Product Overview
The kit is suitable for extracting high purity total DNA from fresh or frozen animal tissues, cells, blood, bacteria and other samples. DNA fragments up to 50 kb in length can be purified without the use of toxic solvents such as phenol or chloroform and ethanol precipitation. An optimized buffer system binds DNA efficiently and specifically from the lysate to the silica-based spin column. PCR and other inhibitors of enzymatic reaction could be effectively removed by two-step washing steps. Finally, high purity DNA could be obtained by using low-salt buffer or water elution. The purified DNA can be directly used in enzyme digestion, PCR, Real-Time PCR, library construction, Southern Blot, molecular markers and other downstream experiments.
The sample is digested with 20 µL of Proteinase K and 200 µL of Solution GS at 56°C for 10 min, then mixed with 350 µL of Solution GA2 and 350 µL of absolute ethanol and loaded onto the adsorption column. The column is washed with 500 µL of Solution RP and 600 µL of Wash Buffer, dried by centrifugation at 12,000 rpm for 2 min, and eluted with 50–200 µL of Solution GE or sterilized water after 2–5 min at room temperature. Purified DNA has an OD260/OD280 of 1.7–1.9.

Storage & Handling
Proteinase K is stored at -20°C for 1 year. Solutions GA1, GS, GA2, RP and GE, the Wash Buffer and the adsorption columns are stored at room temperature for 1 year.
Usage Notes
- Avoid repeated freezing and thawing of samples.
- Add ethanol to the Wash Buffer and Solution RP before use.
- Check the solutions for crystallization and dissolve them at 37°C if needed.
- Confirm that digestion has produced a clear solution before loading the column.
- Remove residual ethanol from the column before elution.
- If eluting with water instead of Solution GE, use water at pH 7.0–8.5.
