Scientific Background
Urea is the primary nitrogen-containing excretory metabolite of protein catabolism, formed in the liver via the ornithine cycle (urea cycle) from ammonium ions generated by amino acid deamination and from arginine metabolism. Blood urea nitrogen (BUN) quantifies the urea fraction of total blood nitrogen; normal adult reference values are 7–21 mg/dL, with BUN:creatinine ratios further distinguishing pre-renal from intrinsic renal causes of azotemia.
In neuroscience and nephrology research, BUN elevation reflects impaired glomerular filtration or augmented protein catabolism associated with systemic stress, sepsis, or corticosteroid administration. Uremic retention solutes—including urea itself—have been mechanistically linked to neuroinflammation, disruption of the blood–brain barrier, and cognitive dysfunction in chronic kidney disease (CKD) patients, a syndrome now termed CKD-associated neurological complications.
Non-invasive BUN measurement in urine or saliva, combined with creatinine normalization, provides a practical tool for monitoring renal metabolic clearance in longitudinal neurological studies without venipuncture.

Assay Overview
This kit uses a direct colorimetric enzymatic reaction for quantitative Urea Nitrogen measurement without the need for sample pre-extraction. A calibrated standard (traceable to NIST reference material where applicable) is provided to construct a standard curve for interpolation. Results are read on a standard microplate spectrophotometer.

This colorimetric assay is validated for use with: Serum, Plasma, Urine, Saliva, Tissue Culture Media. Samples should be diluted with the supplied Assay Diluent or deionized water as specified in the protocol. Lipemic, highly pigmented, or protein-rich samples may require additional dilution to minimize matrix interference.
The kit has a sensitivity of 0.042 mg/dl and a quantification range of 0.156–10 mg/dl. This range is appropriate for typical physiological concentrations. Samples outside the range should be diluted (upper) or concentrated using ultrafiltration (lower).
Total assay time including incubation is approximately 30 Minutes. Refer to the Kit Booklet for the detailed protocol and timing requirements.
The kit contains: Clear 96-Well Plates, Urea Nitrogen Standard, Color Reagent A, Color Reagent B, Urease Enzyme. All reagents are pre-formulated and lot-matched. A calibrated creatinine or nitrogen standard is included for standard curve generation.
A standard 96-well microplate spectrophotometer capable of reading absorbance at the target wavelength (typically 490–550 nm for colorimetric reactions; see Kit Booklet for exact wavelength) is required. The assay is not compatible with a fluorescence reader.
Urine and tissue culture supernatant samples should be diluted with deionized water or assay diluent as specified in the protocol (typically 1:20 for urine). Serum/plasma samples may be used undiluted or at minimal dilution. No extraction step is required. Sample creatinine values are reported in mg/dL; normalize to urine volume or creatinine concentration as appropriate for your experimental design.
