Compound Overview
bpV(phen), an insulin-mimetic agent, is a potent inhibitor of protein tyrosine phosphatases (PTP) and PTEN, with IC50 values of 38 nM, 343 nM and 920 nM against PTEN, PTP-β and PTP-1B, respectively. It suppresses proliferation of the protozoan parasite Leishmania in vitro, drives strong secretion of numerous chemokines and pro-inflammatory cytokines, and activates a Th1-type pathway (IL-12, IFNγ). It can also induce cell apoptosis and shows anti-angiogenic and anti-tumor activity[1][2][3][4][5]. It is supplied as a light yellow to yellow solid (C12H8KN2O5V, MW 350.24) at 98.0% purity.

In Vitro
In H/R-injured H9c2 cells, bpV(phen) (5 μM; 24.5 hours) further decreases cell viability[1]. bpV(phen) (5 μM; 24.5 hours) raises apoptosis in H/R-injured H9c2 cells[1]. Cytoplasmic Cytochrome C accumulates significantly more in H/R-injured H9c2 cells treated with bpV(phen) (5 μM; 24.5 hours)[1]. PTEN-induced putative kinase protein 1 (PINK1)/Parkin-mediated mitophagy is inhibited following bpV(phen) stimulation[1]. bpV(phen) acts as an insulin mimetic after insulin-receptor tyrosine kinase hyperphosphorylation and activation[4].
