Description
Firefly luciferase is a protein with a molecular weight of approximately 61 kDa. In the presence of ATP, magnesium ions, and oxygen, it can catalyze the oxidation of luciferin to oxyluciferin, during which a bioluminescence with a wavelength of around 560 nm is emitted.

This kit is compatible with the vast majority of mammalian cell lines and primary cells, including HEK293, HeLa, Jurkat, CHO, and NIH/3T3. The included cell lysis buffer achieves complete lysis of both adherent and suspension cultures. Yeast or bacterial cells are not recommended without protocol adaptation.
A luminometer or microplate reader with a luminescence detection mode is required. Standard fluorescence readers with excitation sources are not suitable. Compatible instruments include the Promega GloMax, BioTek Synergy, PerkinElmer EnVision, or equivalent luminometers. Ensure any injection system is primed before measurement to minimize signal delay.
Luciferase substrates are light-sensitive and mildly temperature-sensitive. Handle under subdued lighting. After preparing working dilutions, use within the timeframe specified in the datasheet (typically 30–60 minutes). Store concentrated substrate stocks at –20°C in amber or foil-wrapped tubes. Do not subject substrates to repeated freeze-thaw cycles.
The linear detection range exceeds 8 orders of magnitude of enzyme concentration, making this kit suitable for detecting both weak promoter activity and highly expressed reporter constructs without signal saturation. This wide dynamic range is particularly valuable for dose-response and titration experiments.
