This product is a type II restriction endonuclease, a recombinant protein encoded by the BspQI gene from Bacillus sphaericus expressed in E. coli. It recognizes the sequence 5'-GCTCTTCN1/N4-3' and is mainly used for digesting plasmids to prepare linearized DNA fragments with poly(A/T/G/C) ends, obtaining specific sticky ends.
Applications
Digesting plasmids to prepare linearized DNA fragments with poly(A/T/G/C) ends.
Digesting DNA to obtain specific sticky ends.

Shipping and Storage
The products should be stored at -25℃ ~ -15℃ for 1 year.
Store this enzyme at -20°C and avoid repeated freeze-thaw cycles to preserve catalytic activity. The product is shipped Dry Ice and remains stable for up to one year from the date of manufacture when stored under recommended conditions. Aliquoting the stock solution into single-use volumes is recommended for enzymes used infrequently to minimize thermal cycling of the bulk stock.
Digesting plasmids to prepare linearized DNA fragments with poly(A/T/G/C) ends. Digesting DNA to obtain specific sticky ends.. Always verify compatibility with your specific template, buffer, and downstream workflow.
One unit (U) is defined as the amount of enzyme required to completely digest 1 µg of lambda DNA (or specified substrate DNA) in 60 min at the recommended incubation temperature in a total reaction volume of 50 µL.
This enzyme is produced as Recombinant (E. coli) and supplied as a Research Use Only (RUO) reagent. Each lot is subjected to activity assay, purity assessment by SDS-PAGE, and functional validation prior to release. A Certificate of Analysis (CoA) and Safety Data Sheet (SDS) are available on request.
Restriction enzyme efficiency depends on buffer composition (NaCl/KCl concentration, pH, Mg²⁺), incubation temperature (typically 37°C), and template quality. Star activity—non-specific cleavage at relaxed recognition sequences—can occur under conditions of excess enzyme, extended incubation, high glycerol concentration, or non-optimal ionic strength. Use of manufacturer-supplied buffers and adherence to recommended enzyme:DNA ratios (typically 1–10 U per µg DNA) minimizes star activity.
