Background
BTLA (B and T lymphocyte attenuator, also called CD272 and referred to here as BTLA1) is a co-inhibitory receptor of the immunoglobulin superfamily expressed on T cells, B cells, and other lymphocytes. Unlike most checkpoint receptors, BTLA binds a TNF-family ligand, HVEM, and the resulting interaction recruits phosphatases through ITIM motifs to attenuate antigen-receptor signaling. By dampening lymphocyte activation, BTLA contributes to peripheral tolerance and limits excessive immune responses, but it can also restrain anti-tumor immunity. Because BTLA engagement suppresses NFAT-dependent transcription downstream of the T cell receptor, NFAT reporter systems provide a quantitative readout of BTLA signaling for checkpoint and immunotherapy research.
Product Description & Applications
The BTLA1/NFAT Reporter Lentivirus is a two-component immunotherapy reporter system. A receptor construct constitutively expresses the human BTLA receptor together with an antibiotic selection marker, while a reporter construct drives a dual reporter through tandem NFAT response elements coupled to a minimal promoter. Reporter options include a fluorescent protein (GFP or RFP) and secreted Gaussia luciferase, allowing readout by both fluorescence and luminometry.
Sequential transduction and selection generate a dual-stable effector cell line that responds quantitatively to receptor engagement. Applications include studying BTLA checkpoint signaling, testing checkpoint-blockade agents, and characterizing T cell inhibition. Supplied as VSV-G-pseudotyped lentiviral particles purified by PEG precipitation and sucrose gradient centrifugation, suitable for primary and thawed cells.

This 2-vial immunotherapy reporter system consists of a Vial 1 Receptor Lentivirus encoding human BTLA1 under a constitutive promoter with antibiotic selection, and a Vial 2 Reporter Lentivirus encoding tandem NFAT (or NF-κB) response elements driving a dual reporter (GFP, GFP-P2A-GLuc, GLuc, GLuc-P2A-GFP, GLuc-P2A-RFP, RFP, RFP-P2A-GLuc). Sequential transduction and selection generates a dual-stable effector cell line that responds quantitatively to receptor stimulation with a ratiometric fluorescent + bioluminescent readout.
Secreted Gaussia luciferase (where included) accumulates in conditioned media, enabling kinetic sampling without cell lysis. The combined fluorescent and luminescent outputs allow parallel microscopy-based visualization and plate-reader luminometry from the same cell population — providing assay redundancy and flexibility for potency testing formats compliant with regulatory expectations for cell-based functional assays.
