In Vitro
In HT-29, OVCAR3 and SKBR3, BX-2819 (0.001-10 μM, 16 h) can inhibit ErbB2 protein expression and significantly enhances Hsp70 expression[1]. Proliferation of SKBR3 (IC50=14 nM), SKOV3 (IC50 = 36 nM), 50 MKN45 (IC50=72 nM), and NCI-N87 (IC50= 7 nM) tumor cancer cells can be inhibited by BX-2819 (0.001-10 μM, 72 h)[1]. In HepG2 and RPTEC cells, no significant cytotoxic effects are produced by BX-2819 (100 μM)[1].

In Vivo
In NCI-N87 and SKOV3 tumor mice, ErbB2 protein expression is significantly inhibited by BX-2819 (100 mg/kg, intraperitoneal injection (i.p.), twice a week), and in HT-29 and NCI-N87 tumor mice, tumor growth and final tumor load are significantly reduced[1]. In rats, a single intravenous (i.v.) injection of BX-2819 (2 mg/kg) gives a serum half-life that is relatively short (t1/2=0.8 h), alongside a high clearance rate (CL=159 mL/min/kg) and a large distribution volume (Vss=4.5 L)[1].
