C-646 is a potent and selective inhibitor of the histone acetyltransferase p300/CBP, with a Ki of 400 nM. In neuroscience, C-646 has been shown to enhance synaptic plasticity and memory formation, particularly in fear extinction paradigms. It also promotes tau deacetylation, thereby reducing levels of pathogenic phosphorylated tau (p-tau), a hallmark of Alzheimer’s disease. These properties position C-646 as a promising epigenetic modulator for neurodegenerative disease research, offering insights into chromatin remodeling and transcriptional regulation in neuronal health and disease.
