Assay Principle
This sandwich ELISA quantifies canine interleukin-4 (gene IL4) in cell culture supernatants, serum and plasma (heparin, EDTA). Strips are pre-coated with a capture antibody. After the sample is added, a biotinylated detection antibody binds the captured protein, and an avidin–biotin–peroxidase complex supplies the enzyme. TMB develops color in proportion to the bound target; the reaction is stopped and read at 450 nm, and concentrations are read from the standard curve.
Interleukin-4 (IL-4) is a key type 2 cytokine made by Th2 cells, mast cells, basophils and eosinophils. Signaling through IL-4Rα paired with either the common gamma chain or IL-13Rα1, it activates JAK–STAT6 to drive Th2 differentiation, B-cell class switching to IgE and alternative macrophage activation. It is a core readout in allergy and parasite immunity studies.
