Assay Principle

This kit uses a sandwich ELISA format for the quantitative measurement of 1 Beta (IL1b). A capture antibody coated on the microplate binds the target antigen; a detection antibody conjugated to HRP then generates a colorimetric signal proportional to the concentration of analyte in the sample.

What assay principle does this 1 Beta (IL1b) ELISA Kit use?

This kit uses a sandwich ELISA format. A capture antibody pre-coated on the microplate binds 1 Beta (IL1b) from your sample. A biotinylated detection antibody then recognises a second epitope on the captured analyte, followed by an HRP-conjugated streptavidin step that generates a colorimetric signal. Signal is directly proportional to analyte concentration, and results are read at 450 nm.

Which sample types are compatible with this assay in Canis familiaris; Canine Dog?

This kit has been validated for use with serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. Samples should be diluted appropriately within the assay detection range (15.6-1000pg/mL). Cell culture supernatants and tissue homogenates may require preliminary centrifugation to remove particulates. Avoid repeated freeze-thaw cycles — aliquot samples before freezing and use within the validated stability window. Haemolysed, lipaemic, or heavily precipitated samples may interfere with assay performance.

What is the detection range and sensitivity of this kit?

The detection range is 15.6-1000pg/mL with a minimum detectable concentration (sensitivity) of 6.4pg/mL. Samples with concentrations above the upper limit of the range should be diluted with the provided sample diluent and re-assayed. Samples falling below the lower limit may indicate undetectable levels or may require sample concentration prior to assay. Always run a standard curve in duplicate in every plate for accurate interpolation.

What is the difference between the Traditional and Ready-to-use kit formats?

Traditional format contains concentrated reagents that require preparation prior to use (dilution of wash buffer, detection antibody, streptavidin-HRP, etc.). This format offers maximum flexibility and is preferred by experienced ELISA users who wish to customise incubation conditions or buffer composition. Ready-to-use format provides all working-strength reagents pre-prepared, significantly reducing hands-on time and minimising inter-operator variability. This format is ideal for routine clinical research, high-throughput laboratories, or users new to ELISA. Both formats use the same antibody pairs and deliver equivalent analytical performance.

What are the storage conditions and shelf life for this kit?

Store all kit components at 2–8 °C upon receipt. Do not freeze reagents unless specified in the Instruction Manual, as this may compromise antibody activity and standard integrity. The microplate should be returned to its foil pouch with desiccant and resealed after each use. Shelf life: Traditional: 12 months | Ready-to-use: 16 months from manufacture date when stored under recommended conditions. Refer to the lot-specific Instruction Manual for the exact expiry date printed on the kit label.
