Scientific Background

Cas9 (Streptococcus pyogenes CRISPR associated protein 9) is an endonuclease enzyme that, when recruited to a specific DNA sequence by the sgRNA (single guide RNA), introduces a double stranded break into the DNA. This double stranded break is repaired in mammalian cells either through Non-Homologous End Joining or Homologous Recombination. Non-Homologous End Joining often results in the deletion or insertion of several base pairs at the cut site, which, when resulting in a frameshift, causes the functional inactivation of the targeted gene. Cas9-expressing HEK293 cell lines can be transduced or electroporated with sgRNA targeting a gene of interest to quickly generate knock-out cell pools or cell lines.

Product Description

CRISPR

This cell line was generated by limiting dilution from a cell population stably transfected using Cas9 lentivirus (BPS Bioscience #78066). Isolated clones were screened based on Cas9 expression to obtain a high-expressing cell line. The expressed Cas9 protein includes a C-terminal FLAG tag.

What is the parental cell line for this product?

This product is engineered on a HEK293 background (S. pyogenes origin). The HEK293 host was selected for its compatibility with stable transfection and the target pathway or assay type. Consult the product datasheet for passage number guidance and recommended culture media.

What biosafety level is required for this cell line?

This product is classified as BSL-2. Work must be performed in a certified BSL-2 laboratory by trained personnel. All procedures that may generate aerosols must be conducted inside a Class II Biological Safety Cabinet. Institutional Biosafety Committee (IBC) approval is required before use.

Has this cell line been tested for mycoplasma contamination?

Yes. This cell line has been screened using the MycoAlert™ Mycoplasma Detection Kit (Lonza, #LT07-118) to confirm the absence of Mycoplasma contamination. MycoAlert Assay Control Set (Lonza, #LT07-518) was used as a positive control. We recommend that you independently confirm mycoplasma-negative status after receipt and periodically during routine culture using a validated detection kit.

What are the recommended storage conditions?

Store this product at Liquid Nitrogen. Specifically: Cells will arrive upon dry ice and should immediately be thawed or stored in liquid nitrogen upon receipt. Do not use a -80°C freezer for long term storage. Transfer cells from dry-ice shipping to the recommended storage immediately upon receipt. Avoid repeated freeze-thaw cycles, which reduce viability and may alter expression characteristics.

Is a license required to use this product?

Yes, a license is required (Yes). Purchase of this cell line grants a time-limited research-use license for use in your immediate laboratory only. This license does not permit redistribution, sub-licensing, transfer to other institutions, or commercial use. Refer to the License Disclosure section on this page or contact BPS Bioscience for details regarding modifications or commercial licensing.

What selection markers are present in this cell line?

This cell line contains the following selection marker(s): C-terminal FLAG-tag. The selection marker was used during stable integration to identify and maintain transgene-positive cells. Maintain selection pressure with the appropriate antibiotic during routine culture to preserve transgene expression; refer to the product datasheet for recommended concentrations.
