The Cas9 Lentivirus delivers high-titer particles pre-packaged with a Cas9 expression vector and a fluorescent reporter or drug-selection marker for convenient detection of transduced cells. A P2A-linked design allows separate expression of Cas9 and the marker from one mRNA. Background

Cas9 is an RNA-guided DNA endonuclease derived from the bacterial CRISPR adaptive immune system. When complexed with a guide RNA, Cas9 recognizes a complementary genomic sequence adjacent to a protospacer-adjacent motif and introduces a double-strand break, which the cell repairs through error-prone non-homologous end joining or templated homology-directed repair. This programmability has made Cas9 a foundational tool for targeted gene knockout, knock-in, and editing across virtually all model systems. Stable expression of Cas9 in mammalian cells is often a prerequisite for efficient genome editing and for pooled CRISPR screens, making constitutive Cas9 delivery a widely used research resource.

The Cas9 Lentivirus delivers high-titer lentiviral particles pre-packaged with an expression vector for Cas9 together with a fluorescent reporter or drug-selection marker for convenient identification of transduced cells. A PGK promoter drives Cas9, and the downstream reporter or selection marker is separated by a P2A self-cleaving peptide, allowing expression of two distinct proteins from a single mRNA. ORF cloning accuracy has been confirmed by sequencing, and protein expression has been validated by transient transfection. The product supports stable Cas9 expression for targeted genome editing and CRISPR screening, and efficiently transduces difficult-to-transfect cells, including primary and thawed cells. Supplied purified by PEG precipitation and sucrose gradient centrifugation, with each vial containing 380-500 uL at a total titer of 1x10^6 TU.

About This Product

This ORF cDNA lentivirus enables stable overexpression of Cas9 in mammalian cells via a third-generation, VSV-G pseudotyped delivery system. The ORF cDNA is fused to a C-terminal epitope tag (V5, Myc, or HA) and expressed under a strong constitutive promoter (CMV, EF1a, PGK). Reporter and selection marker components (BFP, Firefly Luc, GFP, RFP; Blasticidin, Bleomycin, Puromycin) are co-expressed via self-cleaving P2A peptides, enabling independent protein production without fusion-tag artifacts.

Ultra-purification by PEG precipitation and sucrose gradient centrifugation yields high-titer particles suitable for primary cells, suspension cultures, and stem cells. Stable polyclonal cell lines are established within 10–14 days by antibiotic selection or FACS sorting. For in vivo applications, the serum-free formulation and VSV-G envelope support direct administration or further concentration for stereotactic injection.
