Caspase-2 Fluorometric Assay Kit

SKU:BHT19600017
Overview
Click light‑blue chips for details
Caspase-2 is the most evolutionarily conserved caspase and acts upstream of mitochondrial permeabilisation. This kit measures the activity of Caspase-2 by fluorescence at Ex/Em = 400/505 nm. Completed in 1-2 hours. Rising caspase-2 activity points to genotoxic or mitotic stress engaging the PIDDosome, upstream of any mitochondrial commitment.

Apoptosis Assay Kit

Pathway Context

Caspase-2 is the most evolutionarily conserved caspase and acts upstream of mitochondrial permeabilisation, recruited to the PIDDosome in response to genotoxic stress and aberrant mitosis.

Assay Principle

The Caspase-2 Fluorometric Assay Kit applies a fluorometric format to this measurement, read at Ex/Em = 400/505 nm. Hands-on and incubation time is 1-2 hours.

The assay is based on the detection of the free AFC fluorescence after cleavage from the labeled substrate VDVAD-AFC (AFC: 7-amino-4-trifluoromethyl coumarin).

Rising caspase-2 activity points to genotoxic or mitotic stress engaging the PIDDosome, upstream of any mitochondrial commitment.

Q.What reader does this caspase assay need?
A.This kit requires a fluorescence-capable plate reader. The readout is taken at Ex/Em = 400/505 nm.
Q.What does this caspase reading tell me about the death pathway?
A.Rising caspase-2 activity points to genotoxic or mitotic stress engaging the PIDDosome, upstream of any mitochondrial commitment.
Q.Why measure Caspase-2?
A.Caspase-2 is the most evolutionarily conserved caspase and acts upstream of mitochondrial permeabilisation, recruited to the PIDDosome in response to genotoxic stress and aberrant mitosis.
Q.What material can I assay?
A.The datasheet does not restrict the assay to a named sample list. Prepare lysates or fluids by your standard route and confirm performance with a spike-recovery check on first use.
Q.How long is the caspase protocol?
A.Combined incubation and handling time is 1-2 hours. Keep this interval identical across plates in the same study, since timing differences show up directly in the endpoint.
Q.How should the kit be stored on arrival?
A.Store at -20°C. Shelf life is 1 year. Unpack promptly rather than leaving the box at the bench, and return components to storage immediately after each use.
