Pathway Context
Caspase-6 is an effector caspase distinguished by its cleavage of lamin A and of huntingtin and APP, which has placed it at the centre of work on axonal degeneration and neurodegenerative disease.
Assay Principle
The Caspase-6 Fluorometric Assay Kit applies a fluorometric format to this measurement, read at Ex/Em = 400/505 nm. Hands-on and incubation time is 1-2 hours.
The assay is based on the detection of the free AFC fluorescence after cleavage from the labeled substrate VEID -AFC (AFC: 7-amino-4-trifluoromethyl coumarin).
Caspase-6 activity is the readout of choice when the question is axonal or neuronal degeneration rather than bulk apoptosis.
