Pathway Context
Different apoptotic stimuli engage different initiator and effector caspases, so profiling the family in parallel from a single lysate is often more informative than assaying any one member.
Assay Principle
The Caspase-Family Colorimetric Assay Kit applies a colorimetric format to this measurement, read at 400-405 nm. Hands-on and incubation time is 1-2 hours.
Caspases cleave a variety of cellular substrates after aspartic acid residues—a characteristic that is central to their role in mammalian apoptosis.
Running the panel in parallel shows which initiator and which effector a treatment engaged, in one lysate.
