C-C chemokine receptor type 2 (CCR2) is a G protein-coupled receptor that plays a pivotal role in regulating immune cell trafficking and neuroinflammatory responses. In the central nervous system (CNS), CCR2 is expressed on monocytes, microglia, and neurons, where it mediates the recruitment of immune cells in response to injury or disease.
CCR2 activation facilitates the infiltration of monocytes into the brain and spinal cord, contributing to both protective and pathological outcomes. In neurodegenerative diseases such as Alzheimer’s, Parkinson’s, and amyotrophic lateral sclerosis (ALS), CCR2 signaling influences microglial activation, cytokine release, and the clearance of neurotoxic proteins. While CCR2-mediated immune responses can support tissue repair and amyloid-beta clearance, chronic activation may exacerbate inflammation and neuronal damage.
The dual role of CCR2 in neuroprotection and neurotoxicity makes it a compelling target for therapeutic intervention. Inhibiting CCR2 signaling may reduce harmful inflammation, while enhancing its function could promote immune-mediated clearance of pathological aggregates.
As a key regulator of immune dynamics in the brain, CCR2 is a promising biomarker and therapeutic target in the development of disease-modifying treatments for neurodegenerative disorders. Its modulation offers a strategic approach to balancing immune surveillance with neuroprotection.
A 1:1000 dilution of SPC-704 was sufficient for detection of CCR2 in 15 µg of rat brain cell lysates by ECL immunoblot analysis using goat anti-rabbit IgG:HRP as the secondary antibody.
