In Vitro

At 60 min, CI-1029 potently inhibits purified HIV-1 protease, at an IC50 of 0.11 nM and Ki of 0.11 nM[1]. In HIV-IIIB-infected CEM cells, CI-1029 gives protection at an EC50 of 0.20 μM, while its cytotoxicity in uninfected CEM cells is low (TC50 of 210 μM), yielding a therapeutic index of 1000[1]. With 30% human serum present, CI-1029 still protects HIV-infected cells, at an EC50 of 6.31 μM[1]. Against multiple protease-resistant HIV strains in PBMCs, CI-1029 stays potent, with a fold increase in EC50 of ≤2 relative to wild-type virus[1].
