Scientific Background

Claudins are integral membrane proteins and major components of tight junction strands which serve as a physical barrier to prevent solutes and water from passing freely through the paracellular space between epithelial or endothelial cell sheets. Claudin-6 is one of the 27 members of the claudin family. Claudin-6 is widely expressed in various tumors but rarely expressed in healthy adult tissues. This cancer specific expression of claudin-6 makes it a promising target as an antigen for cancer immunotherapy. Several therapeutics that target claudin-6 are currently in development or in clinical trials including bispecific antibodies, monoclonal antibodies, and CAR-T cell therapy products.

Product Description

Cell Signaling

Recombinant clonal CHO cell line stably expressing full-length human claudin-6 (CLDN6 gene accession number NM_021195.5). Surface expression of claudin 6 was confirmed by flow cytometry. The stable clonal cell line was selected for high, medium, or low levels of claudin-6 expression compared to the parental CHO-K1 cell line.

What is the parental cell line for this product?

This product is engineered on a CHO-K1 background (Chinese Hamster origin). The CHO-K1 host was selected for its compatibility with stable transfection and the target pathway or assay type. Consult the product datasheet for passage number guidance and recommended culture media.

What biosafety level is required for this cell line?

This product is classified as BSL-1. Standard microbiological practices (gloves, lab coat, eye protection) are sufficient. No specialized containment facility is required beyond a clean bench. Consult your institutional IBC for GMO registration requirements.

Has this cell line been tested for mycoplasma contamination?

Yes. The cell line has been screened to confirm the absence of Mycoplasma species. We recommend that you independently confirm mycoplasma-negative status after receipt and periodically during routine culture using a validated detection kit.

What are the recommended storage conditions?

Store this product at Liquid Nitrogen. Specifically: Cells are shipped in dry ice and should immediately be thawed or stored in liquid nitrogen upon receipt. Transfer cells from dry-ice shipping to the recommended storage immediately upon receipt. Avoid repeated freeze-thaw cycles, which reduce viability and may alter expression characteristics.

Is a license required to use this product?

Yes, a license is required (Yes). Purchase of this cell line grants a time-limited research-use license for use in your immediate laboratory only. This license does not permit redistribution, sub-licensing, transfer to other institutions, or commercial use. Refer to the License Disclosure section on this page or contact BPS Bioscience for details regarding modifications or commercial licensing.

What method was used to generate this cell line?

This stable cell line was generated using Lipofectamine 2000 for transgene delivery into the parental host. The stably integrated cells were selected using the appropriate resistance marker and verified for expression prior to cryopreservation.
