Scientific Background

Creatinine, a non-protein nitrogenous waste product derived from the non-enzymatic breakdown of phosphocreatine in skeletal muscle, is continuously excreted by the kidneys through glomerular filtration with negligible tubular secretion or reabsorption at normal serum concentrations. This makes serum creatinine a cornerstone biomarker of glomerular filtration rate (GFR) estimation and a standard surrogate for renal function in clinical and preclinical settings.

In neuroscience research, serum creatinine measurements inform systemic health monitoring for rodent cohorts exposed to nephrotoxic pharmacological agents or undergoing chronic disease modeling. Elevated serum creatinine in rodent models correlates with accumulation of uremic toxins that potentiate neuroinflammation and contribute to hippocampal and cortical dysfunction.

Its low intra-individual biological variability (<15% CV) makes creatinine an ideal internal reference for multi-timepoint longitudinal studies. Combined with BUN, GFR estimations can assess renal clearance of neurotoxic metabolites in chronic kidney disease and cardio-renal syndrome research.

A standard 96-well microplate spectrophotometer capable of reading absorbance at the target wavelength (typically 490–550 nm for colorimetric reactions; see Kit Booklet for exact wavelength) is required. The assay is not compatible with a fluorescence reader.

Is sample normalization required, and how should samples be prepared?

Urine and tissue culture supernatant samples should be diluted with deionized water or assay diluent as specified in the protocol (typically 1:20 for urine). Serum/plasma samples may be used undiluted or at minimal dilution. No extraction step is required. Sample creatinine values are reported in mg/dL; normalize to urine volume or creatinine concentration as appropriate for your experimental design.
