Overview

Cyclin D1 / CCND1 is the regulatory component of the Cyclin D1-CDK4 (DC) complex that phosphorylates and inhibits members of the retinoblastoma (RB) protein family including RB1 and regulates the cell-cycle during G(1)/S transition. Phosphorylation of RB1 allows dissociation of the transcription factor E2F from the RB/E2F complex and the subsequent transcription of E2F target genes which are responsible for the progression through the G(1) phase. Hypophosphorylates RB1 in early G(1) phase. Cyclin D-CDK4 complexes are major integrators of various mitogenenic and antimitogenic signals. Also substrate for SMAD3, phosphorylating SMAD3 in a cell-cycle-dependent manner and repressing its transcriptional activity. Component of the ternary complex, cyclin D1/CDK4/CDKN1B, required for nuclear translocation and activity of the cyclin D-CDK4 complex. Exhibits transcriptional corepressor activity with INSM1 on the NEUROD1 and INS promoters in a cell cycle-independent manner. [UniProt]

This anti-Cyclin D1 antibody is supplied as Purified (Mouse, Monoclonal (mouse origin), clone 1849CT557.93.5, Mouse IgG1, kappa, Unconjugated) and is designed to support common target-detection workflows after the on-page specifications.

Key elements and design rationale

  • Target: Cyclin D1
  • Format: Purified
  • Species reactivity: Human, Mouse
  • Applications (listed): WB
  • Conjugate: Unconjugated
  • Clone and antibody class: Monoclonal (mouse origin), clone 1849CT557.93.5, Mouse IgG1, kappa

Because antibody performance can depend on epitope context, sample preparation, and biological state, interpret signals using appropriate controls and orthogonal evidence when possible.

Biological background

Cyclin D1 is referenced in public gene/protein resources (e.g., UniProt and NCBI Gene), which provide curated names/synonyms, protein features, and pathway context. When designing assays, consider potential isoforms, post-translational modifications, and cell-type specific expression that may influence observed signal.

Research relevance and current trends

  • Profiling Cyclin D1 expression across model systems, perturbations, and time points to support mechanistic hypotheses.
  • Combining antibody-based detection with multi-omics or imaging readouts to link Cyclin D1 signal with phenotype.
  • Using well-matched controls (isotype controls, genetic perturbations, or independent reagents) to strengthen interpretation of target-associated signal.

Common research applications

  • WB

Use the listed applications as a starting point and tailor experimental design to your sample type and readout requirements.

Notes for experimental interpretation

  • Specificity considerations: closely related family members, isoforms, or PTMs can affect apparent specificity; confirm with independent approaches when critical.
  • Controls: include negative controls and, when feasible, genetic or pharmacologic perturbations to support target attribution in your system.
  • Species and sample context: differences in sequence, expression, fixation, or extraction conditions can change signal behavior across models.
