Enzyme & Substrate
Cytochrome c oxidase is the terminal enzyme of the electron transport chain and the step that consumes essentially all cellular oxygen, so its activity is the most direct enzymatic measure of respiratory capacity.
Assay Principle
The Cytochrome C Oxidase Activity Assay Kit applies a colorimetric format to this measurement, read at 550 nm. Hands-on and incubation time is 45 seconds (initial rate).
This enzyme is probably unique in providing energy for the cell by coupling electron transport through the cytochrome chain with the process of oxidative phosphorylation.
