Compound Overview
D-Fructose 1-phosphate disodium is a key intermediate metabolite in the fructose metabolic pathway. As a signaling molecule linking fructose metabolism to glucose metabolic regulation, it acts as an allosteric modulator that counteracts the inhibitory effect of the glucokinase-regulatory protein complex, finely tuning the direction of hepatic glucose metabolism at the substrate level[1][2]. It is an endogenous metabolite supplied as a white to off-white solid (C6H11Na2O9P, MW 304.10) at 99.75% purity.

In Vitro
| Solvent | Solubility | Notes |
|---|---|---|
| H2O | 200 mg/mL (657.68 mM) | requires sonication |
Aliquot the stock solution and store it at -80°C (up to 6 months) or -20°C (up to 1 month); stored under nitrogen; avoid repeated freeze-thaw cycles.
If water is used as the stock solvent, dilute to the working solution and sterilize it through a 0.22 μm filter before use.

In Vitro
In isolated hepatocytes from fed rats, D-Fructose 1-phosphate disodium stimulates glucokinase activity with a half-maximal effective dose of 30 pmol/mg protein, and glucagon attenuates this stimulation[1]. D-Fructose 1-phosphate disodium (1 mM) is a specific substrate for Clostridioides difficile fructose-1-phosphate kinase, with a Km value of 160 μM, and sulfate and phosphate ions inhibit it[1]. The rat liver GKRP-GCK complex is directly dissociated by D-Fructose 1-phosphate disodium, which relieves the GKRP-mediated inhibition of GCK[2]. In intestinal epithelial cells, D-Fructose 1-phosphate disodium inhibits PKM2, thereby driving HIF-1α activation as well as hypoxic cell survival, villus elongation, and enhanced nutrient absorption[2].
