DB Antigen retrieval solution pH6 is designed for the application in immunohistochemistry to optimize the antibody performance with heat induced antigen retrieval on formalin-fixed, paraffin-embedded tissues.

Procedure
Prepare a working solution by diluting the concentrated DB RETCIT 1:10 in distilled water. The ready to use solution can be used for up to 3 months if stored at 4-8 °C.

pH 6 (citrate-based) retrieval is the most widely used condition and is suitable for the majority of IHC targets on FFPE tissue, particularly surface and cytoplasmic antigens. pH 9 (Tris-EDTA-based) retrieval is better suited for some nuclear antigens and targets that yield poor results at pH 6. Optimal retrieval pH should be empirically determined for each antibody-target combination.
The 10x retrieval solution is compatible with all standard heat-induced epitope retrieval (HIER) methods, including pressure cooker, microwave, water bath, and dedicated automated staining systems. Ensure the tissue sections do not dry during the heating process and allow adequate cooling time before proceeding to blocking and antibody incubation steps.
