DB Antigen retrieval solution pH9 is designed for the application in immunohistochemistry to optimize the antibody performance with heat induced antigen retrieval on formalin-fixed, paraffin-embedded tissues.

Procedure

Prepare a working solution by diluting the concentrate DB RETRIS 1:10 in distilled water. The ready to use solution can be used for up to 3 months if stored at 4-8 °C.

Which antigens benefit most from pH 9 retrieval?

pH 9 (Tris-EDTA) retrieval is particularly effective for nuclear antigens such as Ki-67, p53, histone modifications, and certain transcription factors that are suboptimally retrieved under pH 6 conditions. It also improves retrieval for some cytoplasmic and membrane proteins that have strong crosslinking in the nuclear region. Always empirically test both pH 6 and pH 9 for new antibody-tissue combinations.

Can pH 9 retrieval be used for all tissue types?

While pH 9 retrieval is generally well-tolerated by most tissue types, some delicate tissues (e.g., brain, skin, embryonic tissue) may show section detachment at high temperature with alkaline buffer. Ensure sections are mounted on adhesive-coated slides (e.g., SuperFrost Plus) and allow adequate drying time before deparaffinization. Use a retrieval vessel that allows controlled temperature maintenance.
