DB Detection KIT - rabbit/mouse dual, HRP/DAB is an immunohistochemistry reagent from DB Biotech.
DB Detection kit - rabbit/mouse peroxidase (HRP)/DAB+, is a polymer based system designed for application in immunohistochemistry as a universal kit that can be used with prediluted and concentrated primary antibodies (produced in rabbit and/or mouse) from different manufacturers. It is a two-step detection kit; first the tissue is incubated with the diluted (ready to use) secondary antibody (detection system - detecting both, rabbit and/or mouse IgGs), followed by the incubation of the section with the DAB+ Chromogen solution.

Kit Contents
The kit contains reagents sufficient for 250 tests. There are three separate bottles: | Bottle A: HRP Rabbit/Mouse 50 ml ready-to-use solution containing secondary antibody conjugated with HRP, recognizing specifically rabbit and/or mouse IgG. Presented in buffered solution containing stabilizing protein and preservative. | Bottle B: Substrate Buffer 50 ml – buffered solution (ready to use) containing hydrogen peroxide and preservative. | Bottle C: 2.5 ml DAB+ Chromogen solution. | All reagents (A, B, C) are stable for 24 months when stored at 2-8 °C in dark.
Application Protocol
Substrate working solution: mix 1 ml Substrate Buffer (Bottle B) and 20 µl DAB Chromogen solution (Bottle C). Mix thoroughly. Store at 2-8 °C and apply within 5 days. | After incubating the section with primary antibody and appropriate washing procedure: | 1. Apply the secondary antibody (Bottle A) to the section for 25-30 minutes. | 2. Apply the substrate working solution to the section for 2×3 minutes.

This dual-species detection kit is compatible with both rabbit-derived and mouse-derived primary antibodies. The polymer-based secondary detection system includes anti-rabbit and anti-mouse HRP-linked components, eliminating the need for separate secondary antibodies in routine single-plex IHC workflows.
The kit uses 3,3'-diaminobenzidine (DAB) as the chromogen, producing a brown precipitate at sites of antigen localization. DAB signal is permanent, alcohol- and xylene-resistant, and compatible with standard hematoxylin and eosin counterstaining protocols for tissue visualization.
Antigen retrieval (heat-induced epitope retrieval, HIER) is typically required before IHC on FFPE tissue to reverse formalin-induced crosslinks and restore epitope accessibility. DB Biotech offers compatible antigen retrieval solutions (pH 6 and pH 9) formulated to work with this detection system.
A typical IHC workflow using this detection system takes 3–5 hours including deparaffinization, antigen retrieval, blocking, primary antibody incubation, detection, counterstaining, and mounting. Overnight primary antibody incubation at 4°C is recommended for most targets to maximize sensitivity.
