Diazepinomicin, also known as ECO-4601, TLN-4601, and BU 4664L, is an anticancer and antibacterial agent that can be produced by a Micromonospora strain. It induces apoptosis and inhibits the proteases Rhodesain and Cathepsin L with an IC50 of 70-90 μM, while also showing anti-inflammatory and anti-tumor activity, including activity against hepatocellular carcinoma. The compound displays antiparasitic activity against trypomastigote forms of Trypanosoma brucei (IC50 13.5 μM) and moderate antibacterial activity against certain Gram-positive bacteria (MIC ≈ 32 μg/mL)[1][2]. It is supplied as an off-white to gray solid (C28H34N2O4, MW 462.58).

In Vitro
In hepatocellular carcinoma HepG2 cells, Diazepinomicin (1-25 μM; 24-72 h) has dose-dependent antiproliferative activity[1]. Diazepinomicin shows moderate antibacterial activity against specific Gram-positive bacteria, with an MIC of about 32 μg/mL[2].
