DMPO (5,5-dimethyl-1-pyrroline N-oxide) is a widely used spin trap compound for detecting free radicals and reactive oxygen species (ROS) in biological systems. Its low toxicity and favorable pharmacokinetics make it ideal for in vivo and in vitro studies of oxidative stress—a central mechanism in neurodegenerative diseases such as Alzheimer’s, Parkinson’s, and ALS.
DMPO forms stable nitrone adducts with protein and DNA radicals, enabling precise identification of oxidative modifications that contribute to neuronal damage. These adducts serve as biomarkers for radical-induced injury, offering insights into disease progression and therapeutic response. However, the presence of nitric oxide and related reactive nitrogen species can interfere with DMPO quantification, necessitating careful experimental design.
By enabling real-time detection of oxidative stress at the molecular level, DMPO supports the development of antioxidant strategies and enhances understanding of redox imbalance in neurodegeneration.
A 1:1000 dilution of SMC-189 was sufficient to detect the DMPO nitrone adducts of metmyoglobin when loaded at 100 ng/lane by colorimetric immunoblot analysis using Goat anti-mouse IgG:HRP as the secondary antibody.
