Drug Target Assay Kit

DNA polymerase III synthesizes DNA at the bacterial replication fork, extending the RNA primer made by DNA primase. The alpha subunit is the catalytic subunit of the polymerase, and the kit can be used both to detect bacterial DNA polymerase III activity and for high-throughput screening of bacterial DNA polymerase inhibitors.

The assay conditions are optimized for the bacterial DNA polymerase III alpha subunit. The enzyme is not included and is supplied by the user.

Kit Components

Component DPA100K
10 x Buffer DP 500 µl
100 x DNA template 35 µl
100 x dNTP mix 35 µl
10 x Fluorescence dye 320 µl

Final reaction conditions: 20 mM HEPES pH 7.5, 10 mM Mg(OAc)2, 1 mM DTT, 30 nM DNA, 20 nM DNA polymerase III alpha subunit, 0.1 mM dATP and 0.1 mM dGTP.

Assay Principle

The kit measures the DNA synthesized by the polymerase from a DNA template and dNTPs. After the reaction a fluorescence dye is added and the signal is read at 535 nm (excitation 485 nm); higher polymerase activity gives more signal.

Safety & Handling

For Research Use Only (RUO). Not for use in diagnostic or therapeutic procedures. The DNA Polymerase Assay Kit is shipped frozen. Follow standard laboratory safety practices when handling the reagents. Please contact us to confirm storage conditions.
Q.What does this kit measure?
A.It measures the activity of bacterial DNA polymerase III alpha subunit by detecting newly synthesized DNA with a fluorescence dye. The user adds the polymerase; the kit can also be used to screen polymerase inhibitors.
Q.What is included in the kit?
A.The DNA Polymerase Assay Kit (100 assays) includes 10 x Buffer DP (500 µl), 100 x DNA template (35 µl), 100 x dNTP mix (35 µl) and 10 x fluorescence dye (320 µl).
Q.Is a protocol available?
A.Yes. The manufacturer's protocol (E. coli DNA polymerase Assay, linked under Documents) covers catalog # DPA100K.
Q.How long does the assay take?
A.About 65 min (60 min reaction + 5 min detection). See the protocol for the exact steps.
Q.How should the kit be stored?
A.The DNA Polymerase Assay Kit is shipped frozen. Please contact us for long-term storage conditions.
