Product Overview
This kit uses a unique buffer system and is especially suitable for extracting genomic DNA from fresh plant materials. It is safe and convenient to use. It can remove impurities, proteins and other organic compounds in cells to the greatest extent. This product does not contain harmful chemical components such as phenol, chloroform and so on. It is safer for the operator. The kit yields high-molecular-weight genomic DNA of high purity. The DNA recovered by this kit can be used in various routine operations, including enzyme digestion, PCR, library construction, Southern hybridization and so on.
Plant tissue of 100 mg is ground under liquid nitrogen and lysed in 600 µL of Buffer PGE with 6 µL of RNase A at 60°C for 20 min. The cleared lysate is mixed with 0.3 volumes of absolute ethanol and loaded onto the adsorption column, washed with 500 µL of Solution RP and twice with 600 µL of Wash Buffer, air-dried for 5–10 min and eluted with 30–50 µL of Elution Buffer. Purified DNA has an OD260/OD280 of 1.7–1.9.

Usage Notes
- Avoid repeated freeze–thaw cycles.
- Pre-dissolve precipitated Buffer PGE at 56°C before use.
- Use fresh plant tissue.
- Add absolute ethanol to the Wash Buffer before first use, as instructed in the manual.
- Air-dry the column for 5–10 min at room temperature before elution.
- The dissolved volume affects both yield and final concentration.
