Drug Target Assay Kit

MurA (UDP-N-acetylglucosamine enolpyruvyl transferase) carries out the first committed step of peptidoglycan biosynthesis in bacteria. It moves enolpyruvate from phosphoenolpyruvate (PEP) to UDP-N-acetylglucosamine, forming enolpyruvyl-UDP-N-acetylglucosamine and inorganic phosphate. It is essential and an attractive antibacterial drug target.

Pack Sizes

Both sizes include the E. coli MurA enzyme. The only difference is the number of assays: MURA100KE is for 100 assays and MURA500KE for 500 assays (384-well format), with component volumes scaled.

100 assays (MURA100KE) 100 assays; enzyme included
500 assays (MURA500KE) 500 assays; enzyme included

Kit Components

Component 100 assays (MURA100KE) 500 assays (MURA500KE)
10 x Assay buffer 400 µl 2 ml
100 x PEP 35 µl 170 µl
100 x UDP-N-acetylglucosamine (UGN) 35 µl 170 µl
100 x E. coli MurA (5000 nM) 35 µl 170 µl
Phosphate detection dye (also sold separately as MicroMolar Phosphate Assay Reagent, BHT16300173) 5 ml 25 ml

Assay Principle

The kit measures the inorganic phosphate released when MurA transfers enolpyruvate from PEP to UDP-N-acetylglucosamine with a phosphate detection dye; absorbance at 650 nm is proportional to MurA activity, so inhibitors reduce the signal.

Q.What does this kit measure?
A.It measures the activity of E. coli MurA by detecting the inorganic phosphate released when MurA transfers enolpyruvate from PEP to UDP-N-acetylglucosamine. It is used to screen inhibitors of this bacterial cell wall enzyme and to characterize the enzyme.
Q.What is the difference between the options?
A.Both sizes include the E. coli MurA enzyme. The only difference is the number of assays: MURA100KE is for 100 assays and MURA500KE for 500 assays (384-well format), with component volumes scaled.
Q.Is a protocol available?
A.Yes. The manufacturer's protocol (E. coli MurA Assay, linked under Documents) covers catalog # MURA100KE, MURA500KE.
Q.How long does the assay take?
A.About 65 min (60 min reaction + 5 min detection). See the protocol for the exact steps.
Q.How should the kit be stored?
A.The E. coli MurA Assay Kit Plus is shipped frozen. Please contact us for long-term storage conditions.
