Topoisomerase IV, a complex of ParC and ParE, can relax supercoiled DNA. In this kit, relaxed DNA gives much weaker fluorescence with dye H19 than supercoiled DNA, so the signal falls as relaxation goes on.
Kit Components
| Component | T4RA-100KE |
|---|---|
| 10 x Buffer | 480 µl |
| 10 x Supercoiled plasmid DNA | 405 µl |
| 100 x E. coli topoisomerase IV | 45 µl |
| 10 mM ATP | 420 µl |
| 1500 x Fluorescence dye H19 | 20 µl |
Final reaction conditions: 20 mM Tris-HCl pH 8, 35 mM NH4OAc, 4.6% glycerol, 1 mM DTT, 0.005% Brij35, 8 mM MgCl2, 25 µg/ml supercoiled plasmid DNA, 1 mM ATP and 100 nM topoisomerase IV.
Assay Principle
In the presence of magnesium, relaxed DNA quenches the fluorescence of DNA dye H19 much more than supercoiled DNA does. As topoisomerase IV relaxes the supercoiled plasmid, the fluorescent signal (Ex 485 nm / Em 535 nm) decreases.
