E197 is a DOCK5 inhibitor that disrupts the podosome belt structure of osteoclasts and thereby inhibits bone resorption, with an IC50 of 3.44 μM in human osteoclasts. It also inhibits Rac in DOCK5-expressing HEK293 cells, with an IC50 of 36 μM, and prevents bone loss in mouse ovariectomized models[1]. It is supplied as a white to off-white solid (C19H21ClN2O3S, MW 392.90) at 99.91% purity.
