Compound Overview
Ebselen, also known as SPI-1005, PZ-51, or CCG-39161, is a glutathione peroxidase mimetic and a potent blocker of voltage-dependent calcium channels (VDCC)[1][2]. It potently inhibits Mpro (IC50 = 0.67 μM) and the COVID-19 virus (EC50 = 4.67 μM)[3]. It also inhibits HIV-1 capsid CTD dimerization. As an organoselenium compound, it can permeate the blood-brain barrier and has anti-inflammatory, antioxidant, and anticancer activity[4][5]. It is supplied as an off-white to light yellow solid (C13H9NOSe, MW 274.18) at 99.80% purity.

In Vivo
Choose the formulation that suits the animal model and route of administration; percentages are volume ratios of the final working solution. Start from a clear DMSO stock (see In Vitro above), add the co-solvents one at a time in the order listed, mixing after each addition, and prepare the working solution fresh on the day of dosing. If precipitation or phase separation occurs, gentle warming or sonication can help.

In Vitro
Ebselen (SPI-1005; 0.4-100 μM; 20-24 hours) shows strong antiviral effects at 10 μM in COVID-19 virus infected Vero cells. Ebselen covalently binds C145 of the catalytic dyad in COVID-19 virus Mpro[3]. In the HIV-1 life cycle, Ebselen inhibits early viral postentry events by impairing uncoating of the incoming capsid[4]. In mouse brain, Ebselen crosses the blood-brain barrier and inhibits endogenous inositol monophosphatase. Ebselen also inhibits IMPase (inositol monophosphatase)[5]. QSOX1 enzymatic activity is inhibited by Ebselen, which also suppresses invasion of pancreatic and renal cancer cell lines[6].
