Compound Overview
Emodic acid, also known as NSC624610, is an anthraquinone compound isolated from A. microcarpus that can inhibit cancer cell proliferation by inhibiting NF-κB activity. It can also inhibit phosphorylation of p38, ERK and JNK, the secretion of the tumor-promoting cytokines IL-1β and IL-6, and the expression of VEGF and MMP, thereby suppressing the invasion and migration potential of cancer cells[1]. It is supplied as a brown to red solid (C15H8O7, MW 300.22) at 98.00% purity.

In Vitro
| Solvent | Solubility | Notes |
|---|---|---|
| DMSO | 4 mg/mL (13.32 mM) | requires sonication and warming and heat to 60°C; use freshly opened DMSO (absorbed moisture lowers solubility) |
Aliquot the stock solution and store it at -80°C (up to 6 months) or -20°C (up to 1 month); protect from light, stored under nitrogen; avoid repeated freeze-thaw cycles.

In Vitro
In the murine mammary carcinoma 4T1, MDA-231, and MCF-7 lines, Emodic acid (0-100 μM, 72 h) shows strong antiproliferative activity, with IC50 values of 9.33, 8.56, 7.95 μM, respectively[1]. In the 4T1 cell line, Emodic acid (50 μM, 24 h) can significantly inhibit NF-κB transcriptional activity (by 80.75%), as well as p65 phosphorylation and MMP-9 mRNA expression[1]. Emodic acid (50 μM, 24 h) can inhibit secretion of the tumor-promoting cytokines IL-1β and IL-6 and expression of VEGF, thereby inhibiting invasion and migration of 4T1 cells[1].
