Target ENaC gamma

The gamma subunit of the epithelial sodium channel (ENaCγ) is a vital component of the heterotrimeric ENaC complex, which regulates sodium ion transport across epithelial surfaces. While ENaCγ is traditionally associated with fluid and electrolyte balance in organs such as the kidney, lung, and colon, its emerging role in the nervous system is gaining attention in neurodegenerative disease research.

ENaCγ, in conjunction with the beta subunit, is essential for efficient fluid absorption in distal lung epithelia, particularly under edematous conditions. This functional synergy suggests a broader role in maintaining tissue homeostasis under stress. In the retina, differential expression of ENaC subunits—including ENaCγ—has been observed in models of ocular hypertension, implicating ENaCγ in neuroprotective responses to elevated intraocular pressure, a key risk factor in glaucoma.

Recent insights propose that ENaCγ may influence neural cell volume regulation, glial activation, and blood-brain barrier integrity—processes that are disrupted in neurodegenerative diseases such as Alzheimer’s, Parkinson’s, and multiple sclerosis. Its mechanosensitive properties and role in sodium flux position ENaCγ as a potential modulator of neuroinflammation and ionic imbalance in the central nervous system.

As the field of neuroionics expands, ENaCγ is emerging as a promising target for therapeutic development and biomarker discovery in neurodegeneration, bridging epithelial physiology and neural resilience.

1 µg/ml of SPC-405 was sufficient for detection of gamma-ENaC in 20 µg of rat kidney tissue lysate by colorimetric immunoblot analysis using Goat anti-rabbit IgG:HRP as the secondary antibody.
