Enzyme Activity Assay Kit

Enzyme & Substrate

Enolase catalyses the penultimate step of glycolysis, and its neuron-specific isoform is an established clinical marker of neuroendocrine tumours and neuronal injury.

Assay Principle

The Enolase Activity Colorimetric/Fluorometric Assay Kit applies a dual colorimetric/fluorometric format to this measurement, read at 570 nm (colorimetric) or Ex/Em = 530/590 nm (fluorometric). Hands-on and incubation time is 20-60 minutes.

It catalyzes the dehydration of 2-phosphoglycerate (2-PG) to phosphoenolpyruvate (PEP), in the catabolic glycolytic pathway.

Q.Which reader is required for this activity assay?
A.This kit requires either an absorbance or a fluorescence plate reader. The readout is taken at 570 nm (colorimetric) or Ex/Em = 530/590 nm (fluorometric).
Q.What does the activity measurement tell me?
A.Enolase catalyses the penultimate step of glycolysis, and its neuron-specific isoform is an established clinical marker of neuroendocrine tumours and neuronal injury.
Q.Why measure Enolase Activity/?
A.Enolase catalyses the penultimate step of glycolysis, and its neuron-specific isoform is an established clinical marker of neuroendocrine tumours and neuronal injury.
Q.Which sample types are validated for this enzyme?
A.The datasheet lists cells, serum, other biological samples. Other matrices can usually be adapted, but run a spike-recovery check before relying on the values.
Q.How long does one activity run take?
A.Combined incubation and handling time is 20-60 minutes. Keep this interval identical across plates in the same study, since timing differences show up directly in the endpoint.
Q.How should the kit be stored on arrival?
A.Store kit at -20°C. Shelf life of 24 months. Unpack promptly rather than leaving the box at the bench, and return components to storage immediately after each use.
