Product Overview

EntiLink™ PCR Master Mix (Red) is a ready-to-use conventional PCR master mix solution containing Taq DNA Polymerase, dNTP mix, MgCl2, and optimized buffer system. The amplification can be performed by adding primers and templates during the reaction, which greatly simplifies the operation steps of the experiment. The product contains cresol red, and the PCR product can be directly electrophoresed. This product contains excellent stabilizer and can be stored for 3 months at 4°C. PCR products have 3'-dA overhangs for easy cloning into T-vectors.

A 50 µL reaction is assembled from 25 µL of master mix, 1 µL of each 10 µM primer, a moderate amount of template and ddH2O to 50 µL. Recommended template amounts are 50–200 ng of genomic DNA or 0.1–10 ng of plasmid DNA. Cycling is pre-denaturation at 94°C for 1–5 min, 35 cycles of 94°C for 30 sec, 50–60°C for 30 sec and 72°C for 30 sec/kb (60 sec/kb for gene cloning), and a final extension at 72°C for 10 min.

Q.How should the EntiLink™ PCR Master Mix (Red) be stored and how long is the shelf life?
A.The master mix is stored at -20°C with a shelf life of two years. It contains a stabilizer and can be stored for 3 months at 4°C.
Q.What does the master mix contain and what has to be added?
A.The premix contains Taq DNA Polymerase, dNTP mix, MgCl2, an optimized buffer system and cresol red. Per 50 µL reaction, 25 µL of master mix is combined with 1 µL of each 10 µM primer, template and ddH2O to 50 µL.
Q.How much template should be used per reaction?
A.The manual recommends 50–200 ng of genomic DNA or 0.1–10 ng of plasmid DNA per reaction.
Q.Can PCR products be loaded on a gel directly?
A.Yes. The product contains cresol red, so the PCR product can be directly electrophoresed. Products also carry 3'-dA overhangs for easy cloning into T-vectors.
Q.What annealing and extension settings are recommended?
A.The annealing temperature is set 2–5°C below the theoretical primer Tm and the extension time is 30 sec/kb, or 60 sec/kb for gene cloning. The mix is thoroughly mixed before use.
Q.Is this product for research use only?
A.Yes. The manual states that the product is for research use only.
