Product Overview
EnTurbo™ SYBR Color qPCR SuperMix is an optimized 2× Real-time PCR premix containing HotStarTaq DNA Polymerase, SYBR Green I fluorochrome dye, dNTP and Mg2+. In addition, the balanced K+ and NH4+ ion ratios in the buffer promote specific primer annealing. To ensure a highly sensitive and specific PCR reaction, the reaction can be initiated by simply adding the primer and cDNA template to the ready-to-use PCR master premix. The unique PCR buffer ensures sensitive qPCR on all real-time PCR instruments without optimization.
In addition, the product visualizes the pipetting process through the colour change reaction after the template is added, which greatly improves pipetting efficiency and avoids pipetting errors. A stand-alone 50× ROX Dye is supplied and is added at 0.4–2 µL per 20 µL system depending on the instrument. The blue dye in the mix and the yellow dye in the 10 × Dilution Buffer generate a blue-to-green colour change, and the HotStarTaq DNA Polymerase is activated at 95°C for 30 sec. A 20 µL reaction contains 10 µL of 2× mix, 0.4 µL of each 10 µM primer, 0.4–2 µL of 50× ROX Dye, template and ddH2O, and is run at 95°C for 30 sec, then 35–40 cycles of 95°C for 5 sec and 60°C for 30 sec (two-step; the three-step program uses separate 50–60°C annealing and 72°C extension steps), followed by melt curve analysis.
