Overview

For quantitative determination of HDL and LDL/VLDL cholesterol and evaluation of drug effects on HDL and LDL/VLDL metabolism. The assay uses OD340nm for signal readout. Compatible sample input includes Serum. Typical stated assay timing is 60 min.

Key elements and design rationale

  • Readout format: OD340nm supports plate-based signal acquisition and consistent comparison across matched samples.
  • Sample compatibility: The stated sample scope includes Serum, which is useful when aligning matrix type with calibration and control design.
  • Analytical range context: The supplied specifications include a stated detection limit of 5 mg/dL for interpreting low-signal samples.
  • Feature emphasis: Sensitive and accurate. Requires only 20 µL serum sample. The detection limit of 5 mg/dL, linearity up to 300 mg/dL cholesterol in 96-well plate assay.

Additional feature notes highlight Convenient. Room temperature assay. No 37°C heater is needed; Convenient and high-throughput. Homogeneous “mix-incubate-measure” type assay. Can be readily automated on HTS liquid handling systems for processing thousands of samples per day. Available format information for this listing includes 100 Tests.

Biological background

This product is centered on measurement of hdl and ldl/vldl within the matrices described for the assay. In practice, datasets from this type of format are typically interpreted by comparing relative signal, activity, or abundance across matched control and experimental groups rather than relying on a single value in isolation. Careful alignment of sample matrix, incubation window, and calibration strategy is important when comparing results across plates, operators, or study days.

More details

Cholesterol concentrations in High-Density Lipoprotein (HDL) and Low-Density (LDL)/Very-Low-Density (VLDL) Lipoproteins are strong predictors of coronary heart disease. Functional HDL offers protection by removing cholesterol from cells and atheroma. Higher concentrations of LDL and lower concentrations of functional HDL are strongly associated with cardiovascular disease due to a higher risk of atherosclerosis. The balances between high- and low-density lipoproteins are solely genetically determined but can be changed by medications, food choices, and other factors. Simple, direct, and automation-ready procedures for measuring HDL and LDL/VLDL concentrations are very desirable. BioAssay Systems HDL and LDL/VLDL quantification kit is based on our improved PEG precipitation method in which HDL and LDL/VLDL are separated, and cholesterol concentrations are determined using cholesterol esterase/cholesterol dehydrogenase reagent. In this reaction, NAD is reduced to NADH. The optical density of the formed NADH at 340 nm is directly proportionate to the cholesterol concentration in the sample.

Detection method

Detection limit and analytical sensitivity

Reported detection limit: 5 mg/dL.

Procedures and timing

Stated procedure or timing information: 60 min.

Research relevance and current trends

  • Plate-based quantification and side-by-side group comparison remain central use cases for this assay format.
  • The product notes emphasize multi-sample throughput, making it relevant for screening-oriented and larger batch comparison studies.
  • The description supports intervention-focused study designs in which researchers compare baseline and perturbed conditions.

Common research applications

  • Quantify hdl and ldl/vldl in serum by OD340 nm readout.
  • Compare treatment or phenotype groups using matched serum handling.
  • Monitor time-course or pre/post changes in serum across study conditions.

Interpretation is usually strongest when signal changes are assessed alongside matrix-matched controls, replicate agreement, and the assay's stated analytical window.

Notes for experimental interpretation

  • Matrix composition, background signal, and sample handling can influence apparent response; compare like-with-like whenever possible.
  • Use appropriate blanks, controls, and replicate wells to distinguish biological differences from plate, reagent, or handling variability.
Would you please confirm me cat# EHDL-100 can be used for Rat or not?

Yes, the assay is not species-specific and works with rat serum samples.

Could they be used with lung tissue homogenates?

Yes, but the EnzyChrom™ HDL and LDL/VLDL assay kits were developed for serum samples to detect the various transport forms of cholesterol. For tissue samples I would rather recommend using our cholesterol assay kits ECCH-100/E2CH-100.

We would like to know if, during the procedure and before the OD measurement, the reagent added to the serum inactivates hepathitis C virus.

No. The EHDL-100 assay reagents will not inactivate hepatitis C virus, or any other virus.
