Overview

For quantitative determination of phosphoglucomutase enzyme activity in biological samples. Safe and sensitive. Non-radioactive assay. Linear detection range in 96-well plate: 0.5 to 20 U/L activity. Fast and convenient. The procedure. The assay uses OD460nm for signal readout. Compatible sample input includes Serum, plasma, cell lysate, and other biological samples. Typical stated assay timing is 20 min.

Key elements and design rationale

  • Readout format: OD460nm supports plate-based signal acquisition and consistent comparison across matched samples.
  • Sample compatibility: The stated sample scope includes Serum, plasma, cell lysate, and other biological samples, which is useful when aligning matrix type with calibration and control design.
  • Analytical range context: The supplied specifications include a stated detection limit of 0.5 U/L for interpreting low-signal samples.
  • Feature emphasis: Safe and sensitive. Non-radioactive assay. Linear detection range in 96-well plate: 0.5 to 20 U/L activity.

Additional feature notes highlight Fast and convenient. The procedure involves addition of a single working reagent and incubation for 20 min. Room temperature assay. No 37°C incubator is needed; High-throughput. Homogeneous “mix-incubate-measure” type assay. Can be readily automated to assay thousands of samples per day. Available format information for this listing includes 100 Tests in 96-well plate.

Biological background

This product is centered on measurement of phosphoglucomutase within the matrices described for the assay. In practice, datasets from this type of format are typically interpreted by comparing relative signal, activity, or abundance across matched control and experimental groups rather than relying on a single value in isolation. Careful alignment of sample matrix, incubation window, and calibration strategy is important when comparing results across plates, operators, or study days.

More details

PHOSPHOGLUCOMUTASE(E.C. 5.4.2.2) is an enzyme which catalyzes the interconversion of glucose-1-phosphate (G1P) and glucose-6-phosate (G6P). Phosphoglucomutase (PGM) plays a crucial role in glycogen synthesis and degradation. PGM 1 deficiency can cause a rare congenital disorder of glycosylation. A decrease in PGM activity has also been linked with oxidative stress. In BioAssay Systems’ EPGM-100 assay, PGM converts G1P to G6P, which is then oxidized by glucose-6-phosphate dehydrogenase. The formed NADPH is coupled to a formazan chromogen. The increase in absorbance at 460 nm is directly proportional to phosphoglucomutase activity.

Research relevance and current trends

  • Plate-based quantification and side-by-side group comparison remain central use cases for this assay format.
  • The product notes emphasize multi-sample throughput, making it relevant for screening-oriented and larger batch comparison studies.
  • Short assay timing and plate compatibility support time-course or repeated-measure collection plans when handling is kept consistent.

Common research applications

  • Quantify phosphoglucomutase in serum, plasma, cell lysate by OD460 nm readout.
  • Compare treatment or phenotype groups using matched serum, plasma, cell lysate handling.
  • Monitor time-course or pre/post changes in serum, plasma, cell lysate across study conditions.

Interpretation is usually strongest when signal changes are assessed alongside matrix-matched controls, replicate agreement, and the assay's stated analytical window.

Notes for experimental interpretation

  • Matrix composition, background signal, and sample handling can influence apparent response; compare like-with-like whenever possible.
  • Use appropriate blanks, controls, and replicate wells to distinguish biological differences from plate, reagent, or handling variability.
Does this kit come with phosphoglucomutase?

No, this kit does not come with a phosphoglucomutase positive control.

What was the source of the enzyme used to develop this kit?

Phosphoglucomutase from rabbit skeletal muscle was used when developing this kit.

What cell count should I use when preparing cell lysate?

Our general guideline is to suspend about two million (2 x 10^6) harvested cells in 400 μL PBS on ice, then run a pilot test of the sample at different dilutions to determine the optimal one. When testing this kit with PANC-1 cell lysate, we used 650,000 cells/mL. However, the optimal number may vary depending on your cell line and samples.
