Linear detection range: fluorimetric assay 0.3 - 100 U/L Uricase. Fast. Run time is. The assay uses FL530/585nm for signal readout. Compatible sample input includes Biological samples. Typical stated assay timing is 10 min.
Key elements and design rationale
- Readout format: FL530/585nm supports plate-based signal acquisition and consistent comparison across matched samples.
- Sample compatibility: The stated sample scope includes Biological samples, which is useful when aligning matrix type with calibration and control design.
- Analytical range context: The supplied specifications include a stated detection limit of 0.3 U/L for interpreting low-signal samples.
- Feature emphasis: Sensitive. Use 10 μL samples. Linear detection range: fluorimetric assay 0.3 – 100 U/L Uricase.
Additional feature notes highlight Fast. Run time is 10 minutes for rapid results; Convenient. Room temperature “mix-and-read” procedure can be readily automated for high-throughput assay of thousands of samples per day. Available format information for this listing includes 100 Tests in 96-well plate.
Biological background
This product is centered on measurement of enzyfluo uricase within the matrices described for the assay. In practice, datasets from this type of format are typically interpreted by comparing relative signal, activity, or abundance across matched control and experimental groups rather than relying on a single value in isolation. Careful alignment of sample matrix, incubation window, and calibration strategy is important when comparing results across plates, operators, or study days.
More details
URICASEorURATE OXIDASE (E.C. 1.7.3.3)initiates the oxidation of uric acid, eventually yielding allantoin. Uricase activity is found throughout plants, animals, and bacteria, but it is not found in hominids. As a result, humans develop gout when the levels of uric acid become too high. BioAssay Systems’ method provides a simple and high-throughput assay for measuring uricase enzyme activity. In this assay, uric acid is enzymatically converted to allantoin, releasing H2O2. The resulting H2O2reacts with a specific dye to form a pink-colored product. The change in fluorescence intensity at 530/585 nm is directly proportional to the uricase activity in the sample.
Detection method
Fluorescent (FL 530/585 nm).

Reported detection limit: 0.3 U/L.
Procedures and timing
Stated procedure or timing information: 10 min.
Research relevance and current trends
- Plate-based quantification and side-by-side group comparison remain central use cases for this assay format.
- The product notes emphasize multi-sample throughput, making it relevant for screening-oriented and larger batch comparison studies.
- Short assay timing and plate compatibility support time-course or repeated-measure collection plans when handling is kept consistent.
Common research applications
- Quantify enzyfluo uricase in biological samples by FL530/585 nm readout.
- Compare treatment or phenotype groups using matched biological samples handling.
- Monitor time-course or pre/post changes in biological samples across study conditions.
Interpretation is usually strongest when signal changes are assessed alongside matrix-matched controls, replicate agreement, and the assay's stated analytical window.
Notes for experimental interpretation
- Matrix composition, background signal, and sample handling can influence apparent response; compare like-with-like whenever possible.
- Use appropriate blanks, controls, and replicate wells to distinguish biological differences from plate, reagent, or handling variability.
How do I prepare plant extracts for uricase analysis?
We used a Dounce tissue homogenizer with ice-cold 100 mM Tris-HCl (pH 7.5) and ground up etiolated pea shoots and roots then assayed the extracts fresh. Protease inhibitors can be added, as well.
How do I prepare animal tissue extracts for uricase analysis?
We recommend using exsanguinated tissues then homogenizing in 50 mM Tris-HCl (pH 7.5), 150 mM KCl, 2 mM EDTA with a Dounce homogenizer.
Can I use Optical Density to measure uricase activity with this kit?
Yes, the dye in the kit does have an Optical Density peak at 570nm, but we found that the activity was more linear at lower concentrations of uricase while employing fluorescence. If you choose to use OD, you may need to use a non-linear fit for the standard curve.
