In Vitro
Chelation with 18F-AlF[2] Trapping of 2-10 GBq 18F fluoride in 4 mL water was done on an anion exchange cartridge, followed by elution with 0.30 mL 0.5 M NaOAc pH 3.9. Following incubation of the solution for 5 min at room temperature with 6 μL of AlCl3 in water (10 mM) and 300 μL dimethyl sulfoxide, 20 μL of a FAPI-74 solution (4 mM) was added. The reaction proceeded at 95 °C for 15 min; afterward it was cooled to room temperature, diluted with 5 mL water, and worked up by SPE. Elution of the final product used 0.5 mL ethanol and 5 mL 0.9% saline, and phosphate buffer was spiked in before sterile filtration. For chelation with 68Ga, 1.00 mL 68Ge/68Ga-generator eluate (0.6 M hydrochloric acid; ca. 1.2 GBq) was added to a mixture made up of 15 μL FAPI-74 solution (4 mM in water) together with 310 μL sodium acetate (2.5 M in water) plus 0.50 mL ethanol. PET/CT imaging for tumor volume delineation to guide radiotherapy in lung cancer[3] PANC-1 xenograft mice (9 weeks old, body weight=25.3±1.2 g, n=9) underwent PET imaging on a small animal PET scanner 3 weeks after implantation. Under 2% isoflurane anesthesia, [18F]FAPI-74 (12.4±1.7 MBq) was injected into the tail vein. The bolus injection and the start of the dynamic PET scans (scan duration=70 min, n=2) were simultaneous. Static PET scans (n=7, scan duration=10 min) were acquired 1 h after injection, and a CT scan followed.
