Assay Principle
The Equine Insulin ELISA kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of equine insulin in serum, plasma, and cell culture supernatants. The microplate is coated with mouse monoclonal antibody specific for Insulin. Standards or samples are pipetted into the wells and incubated together with biotinylated anti-insulin monoclonal antibody (Detection A). The insulin present in a sample is bound to the wells by the capture antibody, and then by biotinylated antibody, producing an antibody-antigen-antibody “sandwich”. After washing away unbound biotinylated antibodies, HRP-streptavidin (Detection B) is pipetted to the wells. The wells are again washed, and a TMB substrate solution is added to the wells and exhibits a blue color in direct proportion to the amount of insulin in the initial sample. The reaction is terminated by the addition of an acid, and absorbance is measured at 450 nm. The concentration of insulin in the samples is determined by comparing the O.D. of the samples to the standard curve.
Applications Direct Assays: Insulin concentrations in different tissues.
Fast Equine Insulin ELISA Kit is a Solid-phase sandwich ELISA for the quantitative determination of Insulin. The microplate is pre-coated with a capture antibody specific to the target. Target present in the sample or standard binds the immobilized antibody, is detected with a biotinylated detection antibody and streptavidin-HRP, and quantified colorimetrically against a standard curve.
Kit Components
The kit provides a pre-coated microplate, target standard for standard-curve generation, detection reagents (detection antibody and enzyme conjugate), wash buffer, substrate, and stop solution. Refer to the product datasheet for the complete component list and quantities.

Safety & Handling
This kit is validated for cell culture supernatant, serum, plasma, tissue. Other matrices may require additional in-lab validation.
The total assay time is approximately 3 hours from sample addition to plate reading.
The kit provides a pre-coated microplate, target standard for standard-curve generation, detection reagents (detection antibody and enzyme conjugate), wash buffer, substrate, and stop solution. See the datasheet for the full component list and quantities.
A standard microplate reader capable of measuring absorbance at 450 nm is required for this sandwich ELISA.
