Product Overview
This kit provides a simple, rapid and efficient method for extracting DNA from fresh or frozen tissues. After tissue lysis, the DNA binds to the surface of silica-coated magnetic beads. After rinsing, the high-purity DNA is eluted in buffer TB or deionized water. The purified DNA can be directly used in PCR, Real-time PCR, SNP, STR, next-generation sequencing, pharmacogenomics research and so on. Besides being able to rapidly extract DNA from animal tissues, this kit is also applicable to the extraction of DNA from samples such as blood, cells and bacteria.
The protocol processes 25–30 mg of tissue, with less than 10 mg used for spleen, or 200 µL of blood. Tissue is lysed with Buffer ATL and Proteinase K at 56°C for 30 min, treated with RNase A for 10 min, then bound with Buffer AL, isopropanol and Mag beads PN. The beads are washed with Buffer MB, Buffer DW and twice with 75% ethanol at 1200 rpm for 3 min each, dried for 3–5 min and eluted with 80–100 µL of Buffer TB at 56°C for 10 min. The A260/A280 ratio of pure DNA is 1.7–1.9.

Storage & Handling
All components can be stably stored in a dry environment at room temperature (15–30°C), with a shelf life of 1 year.
Usage Notes
- Mag beads PN must not be frozen or subjected to high-speed centrifugation.
- Redissolve buffers if crystals form before use.
- Gram-positive bacteria require lysozyme pretreatment.
- Use less than 10 mg of sample for spleen tissue.
- The 75% ethanol wash is not listed among the kit components and must be prepared separately.
- Isopropanol is required at the binding step and is not supplied.
