PNGase F is an amidohydrolase that can cleave high-mannose, hybrid, and complex oligosaccharides linked by asparagine, specifically removing N-linked glycans. However, the conventional PNGase F enzymatic reaction often takes several hours to release antibody N-glycans. Due to the preferential release of glycans, incomplete deglycosylation might result in biased outcomes, with the obtained glycan distribution failing to represent the true composition of therapeutic antibodies. Therefore, rapidly obtaining an accurate N-glycan profile is crucial for glycosylation monitoring during the production of antibodies and antibody-fusion proteins used in biotherapeutics.

Fast PNGase F

Store this enzyme at -20°C and avoid repeated freeze-thaw cycles to preserve catalytic activity. The product is shipped Dry Ice and remains stable for up to one year from the date of manufacture when stored under recommended conditions. Aliquoting the stock solution into single-use volumes is recommended for enzymes used infrequently to minimize thermal cycling of the bulk stock.

This enzyme is validated for use in Protein Research applications. It is suitable for use in both standard laboratory research settings and, where applicable, optimized reaction workflows requiring high specificity, sensitivity, or throughput.

One unit (U) is defined as the amount of enzyme that catalyzes the release of 1 µmol of product per minute at 37°C from a defined glycoprotein or oligosaccharide substrate under standard assay conditions.

This enzyme is produced as Recombinant (E. coli) and supplied as a Research Use Only (RUO) reagent. Each lot is subjected to activity assay, purity assessment by SDS-PAGE, and functional validation prior to release. A Certificate of Analysis (CoA) and Safety Data Sheet (SDS) are available on request.

PNGase F cleaves between the innermost GlcNAc and asparagine of all N-linked glycans regardless of glycan complexity. Endo H cleaves only high-mannose and hybrid-type N-glycans at the chitobiose core, enabling differentiation of ER-retained from Golgi-processed glycoproteins. Endo S is an IgG-specific endoglycosidase that cleaves N-glycans from the Fc region of IgG antibodies, useful in antibody engineering and glycan remodeling workflows.
